Always on the lookout for TFF applications in cell therapy processes. Surprised to find this publication on Genetic Engineering & Biotechnology News this morning. More straightforward applications for TFF include cell washing or enabling perfusion in a stir tank format. Stimulation and transduction of T cells is a new one for me! The novel activation method is interesting but seems extremely cumbersome to generate without having much performance benefit over traditional CD3/CD28 bound antibodies. The transduction method via active tangential flow shows marketable improvement over a static method. Only thing lacking is a comparison of a static culture utilizing a reagent like retronectin. Assuming performance evens out with retronectin, TFF would still be advantageous in my opinion. TFF would remove a costly reagent, permit more controlled suspension culture technology, and the possibility to utilize the TFF for perfusion or cell washing. Engineered B cells offer exciting new therapeutic possibilities. The differentiation of B cells into plasma cells results in stable, long-lived and therapeutic levels of protein secretion. Establishing GMP-compatible culturing systems is a pivotal aspect to bringing B cell-derived therapeutic modalities to the clinic. Here, the authors developed a robust culturing method enabling high editing efficiency in these hard to engineer cells. They achieved remarkable expansion factors of over 300-fold, while retaining high viability. Furthermore, not all cells need to undergo terminal differentiation in culture to end-stage plasma cells. Indeed, <10% of the plasma cells in culture establish a long-lived niche leading to therapeutically relevant protein secretion, following transfer into recipients. This can occur through negative selection or through differentiation of non-terminally differentiated antibody secreting cells.

LucidQuest Strategic Insights (lqventures.com) >>> Gene&Cell Therapy >> Bayer takes next step in cell therapy, opening new $250M facility in Berkeley: Bayer is easing into the cell therapy space through the opening of its first dedicated manufacturing facility in Berkeley, CA, to support later-stage clinical trials and potentially beyond. The first asset to be developed at the $250 million facility is Bayer-owned BlueRock Therapeutics’ Phase II-ready Parkinson’s disease asset bemdaneprocel. Bayer envisions the site will also support the potential commercial launch of bemdaneprocel, as well as the Big Pharma’s own cell therapy products that have yet to enter the clinic. The 100,000-square-foot facility has capabilities for cell culture, viral transduction, automated filling of cell therapies and product supply for later-stage studies. The Phase II bemdaneprocel trial is expected to recruit in the first half of 2024. Bayer launched its cell and gene therapy program in December 2020 to consolidate all its efforts under one initiative, following its acquisitions of cell therapy company BlueRock in 2019 and gene therapy company AskBio in 2020. The new cell therapy facility is a part of Bayer’s development at its campus in Berkeley, where the company has invested nearly $500 million to develop over the past five years. Bayer has seven cell and gene therapy projects in development, according to a June 30 SEC filing. BlueRock’s bemdaneprocel is its most advanced asset. In June, BlueRock revealed encouraging Phase I data for bemdaneprocel in Parkinson’s disease. A month later, in August, it announced it would focus on bringing bemdaneprocel into Phase II, laying off 50 employees that primarily worked on its immuno-oncology programs. #lucidquest #genetherapy #celltherapy
